3-6 base change
Our group mutated two bases of the original B21 RBS sequence. The third base of the RBS sequence was changed from an A to G in order to form three hydrogen bonds (Watson-Crick bond) with the corresponding C of the 16S rRNA. It is predicted that this change in base will create a stronger and more stable bond. The sixth base of the RBS sequence was changed from a G to a C. This will ideally decrease the number of nucleotides in the ring. Both of these changes are predicted to stabilize the structure while maintaining optimal efficiency.
